I've had an interesting January. I got a temporary student to help me in the lab, and he's been a great help to me. The flip side of this equation is that since I've got someone working for me (who requires a lot of attention) I feel extremely bad slacking off at work while telling him to do things for me. So I haven't been! I've been really productive spending all of my time doing work during the day, and I've been spending most nights with Legionnette - which also means that I haven't really been spending much time at all on SG. So I apologize to anyone who's been feeling neglected. I'm assuming once my student leaves I'll probably go back to slacking off, which means I'll be around here more often.
Anyway, I got a pretty good response from the last time that I posted pictures taken in the lab so I figured I'd post some more now from my recent stint of productivity.
So here you go.
Technical notes: for those who are interested, there are cerebellar cells with inferior olivary explants taken from P2 type 3 neuregulin heterozygotes and left in vitro for 6 days. Stains included FITC-conjugated phalloidin for actin, anti-vesicular glumatate transporter 1 and anti-GFAP both visualized with a rhodamine conjugated secondary antibody. All nuclear staining was done with DAPI present in Vectashield mounting medium.
A (presumably) unipolar brush cell labeled with calretinin:
Another one, this time shown in context with other neurons' nuclei labeled with DAPI in blue:
An inferior olivary explant stained for the vesicular glutamate transporter (with nuclei stained with DAPI in blue):
The same explant but looking at actin staining:
Lest anyone think I'm prejudiced against neurons, here's a GFAP stain which labels astrocytes, again with DAPI in blue:
