First off, I'd like to thank everyone for their interesting replies to my last journal entry, it was fun reading them and hearing what people have to say about modern vs. classical art forms.
What's funny is that the day after I wrote that I went to see this band play whose leader is the former first trumpet for the Metropolitan opera. So he certainly knows his classical music. Well, his new band is anything but. They're "experimental jazz" to put it nicely, and well, it's not very good. I was thinking about my last journal entry while listening to them, and figured I should have included a caveat; while we should all have open minds about modern forms of art, just because they're modern doesn't mean they're good. That's pretty much all I have to say on the subject.
I'm feeling pretty out of it right now. After a crazy post-SG burlesque party on Saturday, after which I got home around 5:30AM to crash, I got up around 11AM yesterday. I needed to come into the lab to do some experiments, and although I didn't especially feel like going , I came anyway. I've been here ever since, which puts me at about 28.5 consecutive hours of electrophysiology. At some point yesterday I decided that I was going to stay until I got some good data. That can take a while. I spent a good, solid 5 hours trying to set up my new perfusion and drug application system, and eventually scrapped the whole thing around 4AM and built a whole new one, which took several hours of tweaking to get just right. Then I got a bunch of recordings, most of which sucked for whatever reason. But juts now I've finished my best recording ever, and I feel like at least part of this marathon was justified. For anyone interested in particulars, I successfully demonstrated nicotine-induced presynaptic facilitation of glutamatergic neurotransmission from the inferior olive to deep cerebellar nuclear neurons in vitro through alpha7 subunit containing nicotinic acetylcholine receptors, and possibly facilitation of GABAergic transmission from an as-yet unidentified presynaptic source. Very cool stuff, I've been trying to get that particular piece of information for.... about 8 months now. Lots of annoying technical hurdles and what not to deal with in between coming up with the problem and getting an answer. Delayed gratification - it's the name of the game in science.
OK, I can feel myself becoming less and less coherent, so I think I'm going to stop here and go home and get some sleep. At least I got to see the sun rise this morning, it's been a long, long time since I've seen that. Hope everyone else is doing well.
