Ugh, I hate Queen street almost as much as I hate Yonge street. My suggestion is to hit up Bloor street, between Spadina and Bathurst. Once there I recommended eating pie from Future's, Poutine from Mel's (and yes, I know the good poutine is in Quebec, so shut the fuck up SGMontreal, Mel's is a Montreal deli/restaurant), and getting lost in Honest Ed's. If you're around on a wednesday or a thursday, stop in at Labrynth for cheap martini's (wednesday) or cheap cocktails (thursday).
Also, Kensington Market is a good place to go too. Lots of good vintage stores, fruit and cheese markets, Uprising the anarchist book store, delicious King's Cafe, and the ever so wonderful Hot Box Cafe where you can actually sit around and smoke pot while drinking smoothies and being served munchie type foods.
Pretty much everything on Queen street now, you'll find in the mall. However it is apparently the place to be and you can buy some cool art off of local artists. Some good things there, Pages (one of my favourite book stores) and Black Market (the downstairs store is better then the upstairs one). Queen street gets much more interesting if you go west of Spadina.
Take FreakPirate's suggestion on checking out Now and Eye for events going on.
Going across to Toronto Island is a good suggestion too. It has the best view of the city. I usually like to pack a picnic and spend an afternoon there, eating, drinking wine, smoking pot and taking pictures.
Don't go to the shoe museum.
Sneaky Dee's has damn good nacho's. (college and bathurst)
Don't eat at Big Fat Burrito in Kensington Market, no matter how good or busy it looks. I got extremely nauseous almost immediately after eating there veggie burrito and puked. I know 3 other people that have the exact same story.
I'd recommend going to Ottawa since it's quite near the quebec side so you'll have a little overview of Quebec and the bars in quebec are open until 3am unless they are really close to the Ontario's border. And well, Ottawa is the capital, after all. Toronto is also a great place to go.
